As said already, vacuum from the pump will be there right after the engine is fired up. If you do not already have one, get a hand vacuum pump / tester and go after the lock system. You will most likely find that the gate lock element is the culprit from what you have described.

The lock system should maintain vacuum for a good long time and allow you several lock/unlock cycles.
