Suddenly, my rear door will not open? door handles both inside and out pull okay but will not open door. What could have gone wrong? How do I go about this...any assist please.

Lift the lock knob at that door,Is it connected? Often lock knobs are unscrewed and the rod drops out. When you unlock the car the rod jams under the door panel.
If this is the case lock the car with the window down and fish around with a paper clip until the rod is visible. Unlock the car and guide the rod out the hole.

No, the lock button is not detached....it goes up and down as it normally would with the others......it's a mystery?
