Shift Knob

Do anyone know how to remove a shift knob on my Sister's 93 500SEL?

Do you mean the gear stick knob? if so then they just unscrew. They are usually put on really tight though!!

Which side do I have to unscrew? Does the engine have to be on? Does it have to be in a specific gear?

The gear knob should unscrew anti-clockwise.

i would recommend having the stick in neutral so its straight up, and have the handbrake firmly on.
