I have a 2000 4 cylinder, is the number#1 cylinder the one closest to the front of the car?

Correct.

Thanks, so it would be #1 starting at the front of the car, the #3 would be second cylinder from the front, #4 would be the third cylinder, and #2 would be fourth or closest to the firewall, would that be correct?

No. The ignition firing order is 1-3-4-2. The physical cylinders are inline from front to back, 1-2-3-4. Cylinder 1 is closest to the front, cylinder 2 is second from front, cylinder 3 in second from rear, and cylinder 4 is at the rear by the bulkhead (i.e. firewall).
