Did you replace the oring on the bolt that holds the filter? Sometimes if it is old it will leak air into the system when tightened again. If you also changed the pre-filter, make certain the o-ring beneath it was re-installed and that the fuel line attaching to it has a good connection.
The only way to prime is via the starter. You could try putting the rear tires up on a 2 x 4 or 4 x 4 to let gravity "help" the lift pump get fuel to the engine. More than 1/2 tank of fuel helps too. If it still wont start, report back which lines you see air in and we'll help you figure out where the leak is at.

yep. 3 times in a row then let it sit an hour and try again.
If it coughs, KEEP cranking until absolutely certain it is running (it wont hurt the starter) or you'll have to start the priming all over again.

If the line from the bottom of the pre-filter has air in it then you have a bad seal at the pre-filter too.
Did you also replace it?

I suspect you didnt fill the filter up sufficiently.
I pre-fill mine to the brim and it generally fires right up, coughs once and then is fine. Glad you got it going.
