Klunking and Frozen rear wheel.

1972 250C

Klunking noise in rear wheel area
Had a flat tire, changed it, went to drive off and the wheel was locked up.

Rocked back and forth and it freed itself. Drove about 2 blocks and it locked again.

What could be the problem?

What is it, and how do I fix it?

Just a thought...
Do you have alloy wheels, and so did you fit a steel spare?

If so, it's possible that you fitted the steel spare wheel with the bolts that came from the alloy wheel, which, as the steel wheel is thinner than the alli one, the bolts go too far through and foul on the brake caliper.
Usually dosn't free off that easy though.
You might have shorter wheel bolts for the steel spare wheel.
