I can't imagine what the hole is, but it seems that plugging it with a bolt or something would serve well.

As far as the rear main goes, the problem is that the bottom half of the seal goes into the rear of the upper oil pan. The further problem is that the upper oil pan doesn't come out without pulling the motor.
