The local MB dealer's Service Department adjusted the door glass on our 1992 300CE and only removed the interior door panel. I would take a closer look at the adjustment nuts that are visible. The Service Department lead tech commented that the glass adjustment is quite tricky ... not a simple adjustment screw with a locknut, but rather a trial-and-error process -- loosening the corresponding nut/adjusting the glass/retightening the nut/activating the window to check the fit until the optimum fit is achieved. I certainly expected that MB would have designed a more sophisticated adjustment mechanism, but not so.
As for the lower door cladding ... I've seen that removed and didn't notice any access holes for adjusting the door glass. The cladding is held on with numerous clips, some of which will likely break if you attempt to remove the cladding. I'd focus on adjusting the glass from the inside because the door cladding R/R will be another headache that you'll wish you had avoided.
