Insert until it bottoms out in the pan.

Level is measured with the car running and in gear. You need to warm the trans up to the correct temperature and check the temp using a HHT or star diagnostic software. If you don't have access to one of those, a infrared thermometer taking a reading of the transmission pan should get the temperature pretty close. I believe level is usually checked at 80C.
