You use regular ATF in the tranny instead of Dextron III? and can you tell the difference?

Can you more precisely define "regular ATF?" Dexron III is no longer licensed by GM, so the oil companies have been using new and creative names to describe the equivalent product.

What car?
It would be good to know what car you have. If you're driving a W123 like me it specifies Dexron II which is what most bottles sold as standard ATF are. Even if you have a newer model that rquires Dexron III I don't think topping off with Dexron II is any great calamity. If you did a fluid change with the wrong stuff I would say drain and replace. It's much cheaper than a premature transmission rebuild.

I don't think Dexron II is sold anymore. Dexron III is backward compatible and is what is commonly used on ours.
