I've made that trip literally DOZENS of times, as recently as a week and a half ago, and have taken pretty much every possible route. The best route is to take I-75 South (taking the I-475 bypass around Macon is optional...doesn't make any difference, really), then take the FL turnpike into Orlando. There's a couple of tolls on the turnpike, but it's a well-maintained road, traffic usually isn't bad, and it's the fastest, most direct way to Orlando.
I'd also like to repeat the previous post's warning about speeding in FL....They'll nail you. They also LOOOOVE to search "suspicious-looking" cars for drugs. Watch your speedo.
