Question M104 ticking sound after work performed

I serviced the transmission and when finished, during engine running to check ATF level I noticed a ticking sound from top end which I don't believe was there before. My question, can damage happen to the valve train if the engine is rotated backwards? I think I got turned around when underneath and when rotating engine to align TC drain plug, went backwards about 20 degrees. I changed the engine oil afterwards to see if clean oil would eliminate problem, no help. Running Castrol 20-50. I read another post that suggested one quart of ATF in engine. Is it time to pull the valve cover? Help!!!
